Fig. 4

Blocking capacity of the inhibitor AKU-005 on rat and mouse enzymes FAAH, MAGL and ABHD6. Data are from ABPP testing of rat and mouse cortical slices. Samples were incubated in vitro for one hour with the vehicle DMSO (basal) and inhibitor AKU-005 (AKU) 100 nM for rat samples and at different concentrations (100 nM, 500 nM, 1 µM) for mouse samples. A AKU (100 nM) strongly inhibited FAAH activity in rat samples (n = 42, p < 0.0001, Wilcoxon signed-rank test). B AKU (100 nM) strongly inhibited MAGL activity in rat samples (n = 42, p < 0.0001, Wilcoxon signed-rank test). C AKU (100 nM) slightly inhibited ABHD6 activity in rat samples (n = 43, p = 0.05, Wilcoxon signed-rank test). D AKU did not reduce FAAH activity in mouse samples (n = 6). E AKU reduced MAGL activity in mouse samples (n = 6, p = 0.0001 at 100 nM, p = 0.031 at 500 nM, p = 0.006 at 1 µM, Wilcoxon signed-rank test). F AKU did not reduce ABHD6 activity in mouse samples at any concentration (n = 5)
